No. Both J10 pads are on the 5V rail. J10 is an "interruption" of the 5V rail put there to check the 5V supply excluding the rest of the circuit. So, if you have a short, or higher voltages, you don't fry the chip.
When done the test, and you have stable 5V, put the jumper permanently on J10.
To test the voltages, 5V and main rails, use L10 as ground point (black probe). This is the amplifier "star ground". If L10 is not yet mounted, use both pads.
Edit:
To test 5V: red probe on right J10 pad, black probe on L10.
In the picture change "primaire" with 1th secondary and "secondaire" with 2nd secondary. The transformer primary is the windings going to mains. Secondaries are the windings (two) with the "transformed" voltage.
